SEE ALSO...

H-Caldesmon Antibody (h-CALD) is a mouse monoclonal IgG1 kappa light chain antibody that detects the H-Caldesmon protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and immunohistochemistry with paraffin-embedded sections (IHCP). Anti-H-Caldesmon antibody (h-CALD) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. H-Caldesmon, also known as high molecular weight Caldesmon, Caldesmon isoform 1, and muscular Caldesmon, is primarily located in the thin filaments of smooth muscle cells, where H-Caldesmon plays a crucial role in regulating cellular contraction, exocytosis, endocytosis, cell movement, and changes in cell shape. This localization is significant because H-Caldesmon interacts with actin and calmodulin, thereby influencing the contractile properties of smooth muscle and contributing to various physiological processes. Notably, while H-Caldesmon is expressed in smooth muscle tumors of soft tissue, H-Caldesmon is absent in myofibroblasts, making H-Caldesmon a valuable marker for differentiating smooth muscle tumors from other neoplasms, such as bone tumors with myoid differentiation and epithelioid mesothelioma versus lung adenocarcinoma.

I am using H-Caldesmon (h-CALD): sc-58703 monoclonal antibody to stain paraffin embedded tissue. Do you recommend antigen retrieval when using this antibody?

Asked by: Trav11
We recommend performing antigen retrieval with sodium citrate buffer (pH 6) and heat. The full protocol can be found here: https://www.scbt.com/scbt/resources/protocols/immunoperoxidase-staining
