GRP 78 Antibody (76-E6) is a rat monoclonal IgG antibody raised against full-length GRP 78 of mouse origin, reactive with mouse, rat, and human proteins, and is suitable for applications such as western blotting (WB) and immunoprecipitation (IP). GRP 78, also known as BiP or HSPA5, is a member of the HSP70 family of heat shock proteins and functions as a molecular chaperone within the endoplasmic reticulum (ER). GRP 78 plays a crucial role in the proper folding and assembly of nascent polypeptide chains, preventing misfolded proteins from aggregating and assisting in their translocation across the ER membrane. This function is essential for maintaining cellular homeostasis and protein quality control, especially under conditions of ER stress caused by factors like hypoxia, nutrient deprivation, or viral infections. GRP 78 is also a key regulator of the unfolded protein response (UPR), a cellular stress response related to the ER that helps cells adapt to adverse conditions by enhancing their protein-folding capacity. GRP 78′s function is important because dysregulation of GRP 78 expression has been implicated in various diseases, including cancer, neurodegeneration, and diabetes. Overexpression of GRP 78 is often observed in tumor cells, where GRP 78 contributes to tumor survival, proliferation, and resistance to chemotherapy by aiding in the management of increased protein synthesis and folding demands. Studying GRP 78 is vital for understanding mechanisms of protein homeostasis, stress responses, and developing therapeutic strategies for diseases associated with ER stress and protein misfolding.

I see the western blot using this antibody gives a double band pattern at expected molecular weight. Are both the bands representing GRP78 and why is that? If only one of them is GRP78, which one is it?

Thank you for your question. HSPA5/BiP/GRP78 Antibody (76-E6): sc-13539 was raised against full length GRP 78 of mouse origin. No epitope mapping has been performed for this antibody as of yet, so its exact binding site is not known at this point.

Hello. Is this antibody suitable for evaluation of expression of cell surface GRP78 and cytoplasmic GRP78? Thank you in advance!

Thank you for your question. This antibody is not currently recommended for IF or IHC(P). We do not currently have any antigen localization data using this antibody. There are a few product citations that we are aware of using this antibody for IF or IHC. We recommend you review those references for any relevant information. For example, PMID's: 32184660, 30221722 and 31767864.

I would like to use your GRP78 Ab conjugated with Alexa 647 for FACS analysis. I would like to know which Isotype do you recommend for your control. And do you have data showing the expression of GRP78 by FACS with your AB?

The GRP 78 Antibody (76-E6): sc-13539 is recommended for detection of GRP 78 of mouse, rat and human origin by WB and IP. We have not yet tested this antibody in flow cytometry. However, this band is not at the expected size of GRP78 but significantly lower. We received this result in two cell lines with endogenous expression of GRP78 (HEK 293 and HCT-116).

Rated 5 out of 5 by from Good result!I used this Ab (1:500, 16 h 4 °C) to blot the GRP78 in SH-SY5Y cells (30 ug total lysate), secondary antibody was mouse (1:1000, 1 h room temp). The band was very clear and strong at 70-75 kDa.
