GRF-1 Antibody (D-11) is a mouse monoclonal IgG2a antibody that detects GRF-1 in mouse, rat, and human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). GRF-1, also known as glucocorticoid receptor DNA-binding factor 1 or p190RhoGAP, is a crucial transcriptional regulator that binds to the promoter region of the glucocorticoid receptor gene, thereby repressing its expression. This repression down-regulates Rho signaling, which is essential for mediating actin cytoskeletal rearrangements and regulating cell cycle events. GRF-1′s function is further underscored by involvement in cytokinesis, where GRF-1 modulates Rho activity through GTPase-activating protein domain, a process intricately controlled by ubiquitination of GTP binding region and subsequent degradation. Additionally, GRF-1 plays a pivotal role in oligodendrocyte differentiation, a process notably disrupted in malignant glioma tumors, suggesting GRF-1 may act as a tumor suppressor. GRF-1 expression is regulated by glucocorticoids, and GRF-1 exists in two isoforms due to alternative splicing, highlighting the complexity and importance in various cellular processes.
