GM130 Antibody (H-7) is a mouse monoclonal IgG2a kappa light chain antibody that detects GM130 of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). GM130 (H-7) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. GM130, a critical cis-Golgi matrix protein, plays a vital role in organizing and maintaining Golgi apparatus function, essential for proper processing and sorting of proteins and lipids within cells. GM130 ′s structure includes a unique amino-terminus that binds to p115 protein, facilitating transport vesicle docking to Golgi membranes, while its carboxy-terminus anchors to the Golgi structure. This dual binding capability maintains Golgi integrity and function, allowing GM130 to participate in vesicle tethering alongside proteins like Giantin. GM130 ′s interactions with p115 and Giantin are essential for transporting materials from endoplasmic reticulum to Golgi, highlighting GM130 ′s importance in cellular trafficking processes. GM130 undergoes post-translational modifications, such as phosphorylation during mitosis, which regulates function and interactions, further underscoring GM130 ′s significance in cellular dynamics.

I purchased this antibody and used it for western blot ( 1:1000) and (1:500) in 2%milk in TBST but didn't work at a ll. I use whole cells lysate of Hela lysate as a positive control. Can you recommend what concentration to use, please?

Thank you for your question. We recommend using the antibody at a starting dilution of 1:200 in 5% nonfat milk in TBST. However, since you have not observed any signal, we would recommend starting at 1:100. We have not tested the antibody in HeLa whole cell lysate so we do not know about the suitability of this cell line as a positive control for GM130. However, we do recommend using either Hep G2 or NIH/3T3 whole cell lysates, or rat liver tissue extract as a positive control. We recommend loading 50 µg protein per lane. Can you recommend a positive control to use with GM130 (H-7): sc-55590 monoclonal antibody for Western blot?

We recommend Hep G2 (sc-2227) or NIH/3T3 (sc-2210) whole cell lysates to use as positive controls as we have Western blot data showing specific detection of GM130 in these cell lines.

What secondary is best to use with GM130 (H-7): sc-55590 monoclonal antibody?

Usati 20 ug di proteine totali. Diluizione primario 1:500 in latte 5% PBS-T (0,1% Tween). Invece clone B-10 SC-55591 Anti-GM130 Antibody ha dato segnale sugli stessi campioni.
