Glutathione reductase Antibody (C-10) is a mouse monoclonal IgG1 kappa light chain antibody that detects Glutathione reductase of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). Glutathione reductase (C-10) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Glutathione reductase, also known as Glutathione reductase mitochondrial precursor, GRase, GSR, or GR, is a crucial enzyme belonging to the class-I pyridine nucleotide-disulfide oxidoreductase family. Glutathione reductase plays a vital role in cellular defense against oxidative stress by maintaining high levels of reduced glutathione in the cytosol, which is essential for detoxifying reactive oxygen species and protecting cellular components from oxidative damage. Glutathione reductase catalyzes the reduction of oxidized glutathione to its reduced form, utilizing NADPH as a cofactor, thereby ensuring a continuous supply of reduced glutathione for various cellular processes. Glutathione reductase can localize to both mitochondria and the cytoplasm, and forms a disulfide-linked homodimer, with an active site featuring a redox-active disulfide bond that is critical for enzymatic function.

Application: Western blot Sample: whole lysate of human hepatocytes from Huh7 cell line Gel running conditions: reduced, denaturing, 12% SDS-PAGE Loading amount: 25 µg protein/lane Transfer: semi-dry, on nitrocellulose membrane (0.45 µm) Blocking agent: 5% milk in TBST buffer, for 1h, RT Primary antibody dilution- 1: 800 in 5% milk, for 16 h at 4°C Secondary antibody: non- Santa Cruz antibody anti-mouse-HRP, 1:10000 dilution in 5% milk, for 1h, RT Detection: ECL substrate (femto sensitivity) Exposure time: 4 sec Bands: specific, at 50 kDa; non-specific bands are absent.
