GLUD1/2 (C-10) is a mouse monoclonal IgG1 kappa light chain antibody that detects GLUD1/2 protein of mouse, rat and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP) and enzyme-linked immunosorbent assay (ELISA). GLUD1/2 (C-10) antibody is available as non-conjugated form and multiple conjugates including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC) and various Alexa Fluor® forms. GLUD1 (glutamate dehydrogenase 1), also known as GDH, GDH1 or GLUD, and GLUD2 (glutamate dehydrogenase 2), also known as GDH2 or GLUDP1, are mitochondrial matrix enzymes belonging to the Glu/Leu/Phe/Val dehydrogenases family. Existing as homohexamers, GLUD1 catalyzes the oxidative deamination of glutamate to α-ketoglutarate and ammonia while GLUD2 is involved in glutamate recycling during neurotransmission. GLUD1 regulates amino acid induced insulin secretion and is allosterically activated by ADP and inhibited by GTP and ATP. Mutations in GLUD1 cause hyperinsulinism-hyperammonemia syndrome (HHS), an inherited condition characterized by high insulin and ammonia levels in blood. GLUD1 may contribute to learning and memory by increasing glutamate turnover. GLUD2 is expressed primarily in testis and retina, with lower expression in brain.

For Western Blot, is it recommended to use denatured or non-denatured conditions with GLUD1/2 (C-10): sc-515542 monoclonal antibody?

Asked by: Germaine
Thank you for your question. We recommend this antibody for use in denatured Western Blot conditions. It has not been validated for use in non-denatured conditions. Please contact our Technical Service Department for further details or inquiries.

Rated 3 out of 5 by from quite wellwb on mouse brain homogenate: 3 bands, 1 aroud at 50kD seems to be specific

Rated 5 out of 5 by from Excellent antibody!I used this antibody for a western blot of rat liver homogenates. It detected only one band at the correct molecular weight, very clear and clean, without any unspecific bands. I started with a 1:1000 dilution of the antibody, but ended up using a 1:5000 dilution. It works great.
