Gephyrin Antibody (G-6) is a mouse monoclonal IgG2b kappa light chain antibody that detects Gephyrin protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). Anti-Gephyrin antibody (G-6) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Gephyrin plays a crucial role in postsynaptic localization of inhibitory glycine receptors, which are essential for maintaining inhibitory neurotransmission in the central nervous system. Gephyrin is predominantly expressed in the brain, where Gephyrin localizes to neuronal postsynaptic membranes, facilitating the anchoring of glycine receptors to subsynaptic microtubules. Proper functioning of Gephyrin is vital for synaptic stability and plasticity, making Gephyrin a key player in modulating synaptic strength and overall balance of excitatory and inhibitory signals in the brain. At least five splice variants of Gephyrin have been identified in rat and human brain tissue, each potentially contributing to diverse functional roles in neuronal signaling.

I came across that some of postsynaptic proteins, including gephyrin, required milder fixation for IHC (e.g., 1-2 h 2-4% PFA fixation). If I use this antibody (G-6, sc-25311) for IHC of mouse brain sections, will it be essential to do so?

How much protein do you recommend loading on the gel for Western blot detection using Gephyrin (G-6): sc-25311?

IF: Epifluorescent analysis primary hippocampal neurons at day in vitro 15 using Gephyrin (sc-25311; 1:50) (red), GFP (green) and MAP2 (blue). Fixing in 4% PFA + 4% sucrose in PSB for 15 minutes followed 100% methanol post-fixing for 10 minutes at -20*C. WB: Analysis of Gephyrin from P30 mouse brain lysate. Mouse anti-Gephyrin (sc-25311) was diluted 1:1000 in TBS 1x containing 5% skimmed milk and 0,2% Tween-20. Anti-mouse HRP-conjugated secondary antibody was diluted 1:10000.
