SEE ALSO...

GAPDH Antibody (D-6) is a mouse monoclonal IgG2b kappa light chain antibody designed to detect glyceraldehyde-3-phosphate dehydrogenase (GAPDH) across multiple species, including mouse, rat, human, equine, canine, bovine, and porcine, through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). Anti-GAPDH antibody (D-6) targets a peptide located near the C-terminus of human GAPDH. GAPDH is a pivotal enzyme in the glycolytic pathway, catalyzing the conversion of glyceraldehyde-3-phosphate to 1,3-bisphosphoglycerate, which is essential for cellular energy production. Beyond its central role in metabolism, GAPDH is involved in a variety of non-glycolytic functions, including membrane fusion, microtubule bundling, DNA repair, and the regulation of apoptosis. These diverse functions make GAPDH a critical player in numerous cellular processes, including cellular metabolism, cancer cell proliferation, and response to hypoxic conditions. GAPDH interacts with Akt, where phosphorylation by Akt enhances glycolytic activity, thereby supporting cancer cell survival under stress. Additionally, GAPDH interacts with hypoxia-inducible factor 1-alpha (HIF-1α), which increases glycolysis in low-oxygen environments, a key mechanism in tumor progression. GAPDH monoclonal antibody (D-6) is available in both non-conjugated form and various conjugated forms, providing versatility for diverse experimental needs and facilitating advanced research into the multifaceted roles of GAPDH in cellular metabolism and disease mechanisms.

Hi, Why are two bands showing up for GAPDH in some of the example images but not others? Is there a way to optimize how many bands show up?

Asked by: Is17
Thank you for your question. The antibody expresses multiple isoforms but is commonly found around 37KDa. PMID: 1924305 (37) https://www.uniprot.org/uniprotkb/P16858/entry#sequences

buenas tardes, me preguntaba si hay alguna referencia para justificar el protocolo que la suma de las dos bandas se puede utilizar para cuantificar, si ya decidí y tengo los datos de GAPDH como control de carga.

Asked by: KARINA D
Gracias por su pregunta. Desafortunadamente, no tenemos un protocolo para la cuantificación de proteínas en Western Blot.

What is the identity of the two bands? Whether these two bands are isoform?

QUE BANDA ES LA QUE SE CUANTIFICA LA DE ABAJO O LA DE ARRIBA?

Asked by: KARINA D
Thank you for your question. GAPDH has two isoforms, both of which may be present in a given sample. Total GAPDH levels are the sum of both bands produced. If you require an antibody against a housekeeping gene for quantification you could opt for an antibody specific against tubulin or actin.

I am working with zebrafish samples. Do you know if this antibody will cross-react?

Asked by: Cweed
Please contact our Technical Service Department and we would be happy to perform sequence analysis and review our data to answer any questions you may have about additional species reactivity.
