GAP-43 Antibody (B-5) is a mouse monoclonal IgG1 kappa light chain antibody that detects GAP-43 protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). GAP-43 Antibody (B-5) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. GAP-43, also known as growth-associated protein 43, plays a vital role in neuronal development and regeneration, being predominantly expressed in neuronal growth cones during these processes. GAP-43 presence is crucial for the formation of synaptic connections and the overall regenerative response in the nervous system. GAP-43 is involved in the modulation of synaptic plasticity, which is essential for learning and memory. GAP-43 interaction with specific RNA-binding proteins, such as the neuron-specific Elav/Hu family member HuD, enhances the stability of GAP-43 mRNA, thereby increasing GAP-43 levels in developing neurons. This regulation is particularly important, as studies have shown that heterozygous GAP-43 knockout mice, which exhibit a 50% reduction in GAP-43 levels, demonstrate significant impairments in memory tasks, highlighting GAP-43′s critical role in cognitive functions.

Can this mouse monoclonal antibody be used in IF or IHC on mouse tissue without background or non-specific staining issues?

Thank you for your question. Yes, it can be used successfully for this application. We recommend using a directly conjugated mouse monoclonal primary antibody. This mouse monoclonal antibody GAP-43 (B-5) is currently available conjugated to HRP, PE, FITC, AF488 and AF647.

There was medium detection of the ~43 kDa target protein in mouse tissues (brain/testes/thymus/muscle mix). No major cross-reactivities were observed at 2 µg/ml antibody concentration with off-target proteins in the mouse tissues tested.
