GalNAc-TL5 Antibody (F-5) is a mouse monoclonal IgG2a kappa light chain antibody that detects GalNAc-TL5 of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). Anti-GalNAc-TL5 antibody (F-5) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. GalNAc-TL5, also known as GALNTL5, is a crucial member of the UDP-N-acetyl-alpha-D-galactosamine:polypeptide N-acetylgalactosaminyltransferase family, which plays a vital role in initiating mucin-type O-linked glycosylation within the Golgi apparatus. This 443 amino acid single-pass type II membrane protein is specifically localized to the Golgi apparatus and is expressed predominantly in the testis, where GalNAc-TL5 utilizes manganese and calcium as essential cofactors for enzymatic activity. GalNAc-TL5 is responsible for transferring an N-acetyl-D-galactosamine residue to serine or threonine residues on protein receptors, catalyzing the initial step in the biosynthesis of O-linked oligosaccharides. Unlike other members of the GalNAc-T subfamily, GalNAc-TL5 lacks a C-terminal ricin B-type lectin domain, which may influence functional specificity. GalNAc-TL5 features two conserved domains within its glycosyltransferase region: the N-terminal domain, known as domain A or GT1 motif, is likely involved in manganese coordination and substrate binding, while the C-terminal domain, referred to as domain B or Gal/GalNAc-T motif, is thought to play a role in catalytic reactions and UDP-Gal binding. GalNAc-TL5 exists in two alternatively spliced isoforms, further contributing to functional diversity.
