    Overview

    Lgals4 encodes the β-galactoside–binding lectin galectin-4, a tandem-repeat galectin enriched in intestinal epithelium that associates with glycosylated membrane proteins and glycolipids to organize apical microdomains and regulate cell–cell and cell–matrix interactions. Galectin-4 contributes to epithelial differentiation, barrier maintenance, and trafficking of glycoproteins, and can modulate signaling and immune communication through carbohydrate-dependent binding at the cell surface. In mouse tissues, altered Lgals4 expression has been linked to changes in mucosal inflammation and epithelial homeostasis, making it relevant for studies of gut physiology and inflammation-associated remodeling. Its lectin activity also supports investigation of glycosylation-dependent pathways that influence adhesion, migration, and epithelial stress responses.

    galectin-4 CRISPR/Cas9 KO Plasmid (m2) is a pool of plasmids designed for targeted disruption of the Lgals4 gene in mouse cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the Lgals4 together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.

    The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the Lgals4 open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish galectin-4 protein expression.

    This CRISPR knockout system enables efficient generation of Lgals4-deficient cell models for investigation of galectin-4 signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
