    galectin-4 Antibody (E-2) is a mouse monoclonal IgG1 kappa light chain antibody that detects galectin-4 of mouse origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). galectin-4 (E-2) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Galectins, a family of soluble β-galactoside-binding lectins, play crucial roles in modulating cell-to-cell adhesion and interactions with the extracellular matrix, which are vital for processes such as tumor progression, pre-mRNA splicing, and apoptosis. Specifically, galectin-4, also referred to as Gal-4, L36, or LGALS4, is located on human chromosome 19q13.2 and consists of 323 amino acids, featuring two homologous carbohydrate recognition domains (CRDs) that are essential for binding activity. The structural integrity of these domains is critical, as they facilitate galectin-4′s ability to interact with glycoproteins and glycolipids, influencing cellular signaling pathways and adhesion processes. Notably, galectin-4 expression is associated with the malignant potential of human hepatocellular carcinoma and is modulated by factors such as cell-cell contact, serum growth factors, and the differentiation status of cells. In terms of localization, galectin-4 is predominantly expressed in the epithelial cells of the colon, rectum, and intestine, as well as in the HT29 and LS174T cell lines, where underexpression in colorectal cancer and preferential upregulation in cells susceptible to peritoneal dissemination highlight galectin-4′s potential as a biomarker for cancer progression.

    Thank you for your question. You can find our recommended protocol by following this link: https://www.scbt.com/de/resources/protocols/immunoperoxidase-staining. We recommend Heat treatment: Place slides in a container and cover with 10 mM sodium citrate buffer, pH 6.0 (sc-294091); or with 50 mM glycine-HCl buffer (sc-29096), pH 3.5, with 0.01% (w/v) EDTA (sc-29092). Heat at 95° C for 5 minutes. Top off with fresh buffer and heat at 95° C for 5 minutes (optimal incubation time may vary for each tissue type). Allow slides to cool in the buffer for approximately 20 minutes. Wash in deionized H2O three times for 2 minutes each. Aspirate excess liquid from slides.
