frizzled-6 Antibody (C-12) is a mouse monoclonal IgG2b kappa light chain antibody that detects frizzled-6 protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). anti-frizzled-6 antibody (C-12) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. frizzled-6, also known as frizzled homolog 6 in Drosophila, plays a crucial role as a seven-transmembrane domain receptor that binds Wnt ligands, which are essential for regulating cell signaling pathways involved in development and tissue homeostasis. frizzled-6 is particularly important in influencing macroscopic hair patterning and other tissue polarity events, which are vital for proper organ formation and function. frizzled-6 contains a signal peptide and a cysteine-rich domain in its N-terminal extracellular region, but notably lacks a C-terminal PDZ domain-binding motif, which differentiates frizzled-6 from other frizzled family members. frizzled-6 interacts with frizzled-3 to influence neural tube closure and the orientation of hair bundles in auditory and vestibular sensory cells, highlighting frizzled-6′s significance in sensory organ development. In vitro studies using Madin-Darby canine kidney (MDCK) cells have shown that frizzled-6 forms a complex with Wnt-4 through its cysteine-rich domain, further underscoring frizzled-6′s role in cellular signaling. frizzled-6 expression is observed as a 4.4 kb mRNA in various human tissues, including heart, brain, placenta, lung, liver, skeletal muscle, kidney, pancreas, thymus, prostate, testis, ovary, small intestine, and colon, indicating frizzled-6′s widespread importance in human physiology.

We saw a strong band just below the 50 kD marker in HCT116 whole cell extract, but no clear band above it with 1:500 dilution from your sample antibody by western blot. What concentration did you use to detect Frizzed6 protein in HT29 cells?

Thank you for your question. frizzled-6 (C-12) is recommended for detection of CMAS of mouse, rat, and human origin by western blotting (starting dilution 1:100, dilution range 1:100-1:1000), immunoprecipitation [1-2µg per 100-500 µg of total protein (1 ml of cell lysate)], immunofluorescence (starting dilution 1:50, dilution range 1:50-1:500), and solid phase ELISA (starting dilution 1:30, dilution range 1:30-1:300).
