SEE ALSO...

FPR2 Antibody (GM1D6) is a mouse monoclonal IgG1 kappa light chain antibody that detects FPR2 protein of human origin by western blotting (WB), immunoprecipitation (IP), and flow cytometry (FCM). Anti-FPR2 antibody (GM1D6) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. The N-formyl peptide receptor (FPR) family, which includes FPR2, is composed of three members: FPR, FPR-like 1 (FPRL1), and FPR-like 2 (FPRL2). These receptors are integral to the immune response, functioning as chemotactic G protein-coupled receptors with seven transmembrane domains, predominantly located on the surface of phagocytic leukocytes such as neutrophils and monocytes. FPR2 plays a crucial role in mediating inflammatory responses and host defense mechanisms by binding to various ligands, including lipoxin A4 and N-formyl-methionyl-leucyl-phenylalanine (fMLP). Upon activation, FPR2 triggers cellular migration and calcium mobilization, which are vital for effective immune responses. Notably, FPR2′s interaction with specific ligands is essential for its function, as FPR2 helps regulate inflammation and may influence the progression of diseases such as Alzheimer′s. The ability of FPR2 to modulate immune cell activity makes FPR2 a significant target for therapeutic interventions aimed at controlling inflammation and enhancing host defense.

Can FPR2 (GM1D6): sc-57141 monoclonal antibody be used in IF or IHC with mouse tissue or cells? Will there be non-specific staining?

Asked by: Randy McDonald
Thank you for your inquiry. The use of mouse monoclonal antibodies with mouse samples is very common and typically poses no problem. To eliminate any potential cross-reactivity of an anti-mouse secondary detection reagent, FPR2 (GM1D6): sc-57141 is available in a variety of direct conjugations, such as HRP, PE, FITC and AlexaFluors.

Rated 4 out of 5 by from Quite good antibodyWe tested FPR2 by western blotting analysis. 60 micrograms of mouse brain membrane lysates were resolved on 10% SDS-PAGE, and incubated over night with monoclonal FPR2 antibody (1:1000). Secondary antibody utilized was anti-mouse sc-2005 antibody (1:5000). FPR2 signal was detected by chemidoc (Biorad) and the signal (not fully clear) was reached after medium exposition.

Rated 4 out of 5 by from The antibody worked pretty good on freshlyThe antibody worked pretty good on freshly isolated human monocytes and granulocytes, when blocked with 5% heat-inactivated FCS. At a dilution of 1/1000 I observed clear stainings with minimal background and no signal for the isotype control. Tests for cross reactivity in HEK cells expressing hFPR1 or hFPR3 were negative. For high-quality pictures cell had to be fixated with 3% methanol-free paraformaldehyde. In summary the antibody works quite good with good specificity so far.
