FOXN1 Antibody (E-3) is a mouse monoclonal IgG2a kappa light chain antibody that detects FOXN1 of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). FOXN1 (E-3) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. FOXN1, also known as Transcription factor winged-nude (WHN), plays a crucial role in the development of the thymus and skin by regulating the growth and differentiation of epithelial progenitor cells into specialized thymic epithelial cells. This function is vital for proper thymus formation, which is essential for T cell development and immune system function. Mutations in FOXN1 can lead to severe immunodeficiency, as seen in nude mice and rats, which are valuable models for cancer research due to compromised immune systems. FOXN1 expression is primarily active in the skin and thymus, highlighting its importance in these tissues′ development. Additionally, secreted Wnt glycoproteins regulate FOXN1 transcription in the thymus, further emphasizing the intricate signaling pathways involved in regulation. In humans, FOXN1 is expressed at low levels in the kidney and thyroid gland, as well as in differentiating cells of the hair follicle precortex, indicating broader biological significance beyond the thymus.

Are there any issues with using sc-271256: FOXN1 (E-3) mouse monoclonal primary antibody on mouse samples?

To eliminate any potential cross-reactivity of an anti-mouse secondary detection reagent,we recommend using a directly conjugated primary antibody. Dharmacon smartpool siFOXN1 partially reduced the intense band. 15µg MCF10A cell lysate, 4-12% gel, 1hr block (5%milk/TBST). Primary antibody,1:500 overnight (5%milk/TBST). Secondary antibody Invitrogen donkey anti mouse 1:500 1hr. Membrane scanned using the Li-cor CLx Odyssey system.
