FOXE3 Antibody (D-6) is a mouse monoclonal IgG1 antibody that detects FOXE3 in mouse, rat, and human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). FOXE3 is a 288-amino acid winged-helix transcription factor that plays a pivotal role in the early stages of lens development, particularly in the closure of the lens vesicle. FOXE3 not only promotes the survival and proliferation of lens epithelial cells but also prevents their differentiation, ensuring proper formation of the lens. As lens fiber cells begin to differentiate, FOXE3 expression is restricted to the undifferentiated cells on the anterior surface of the lens, highlighting FOXE3′s importance in maintaining the lens′s structural integrity during development. Mutations in the FOXE3 gene can lead to congenital primary aphakia, a rare condition characterized by the absence of a lens, typically arising from a null mutation during the fourth or fifth week of human embryogenesis. Anti-FOXE3 antibody (D-6) is an essential tool for researchers studying lens development and related disorders, providing insights into the molecular mechanisms underlying these processes.

Why does the blot show two different bands or could you precise what does each band correspond to ?

Thank you for your question. The western blot image shows cell lysates that were transfected with a truncated peptide corresponding to the antibody's binding site. Endogenous FOXE3 will produce a signal at 33 kDa in western blots.

Why does the blot show two bands, and neither of which is at 33 kDa (which is the predicted protein size for human FOXE3 protein)?

Thank you for your question. The Western blot data may not be representative of the endogenous molecular weight for FOXE3. This is detected in a human FOXE3 transfected 293T lysate. The observed molecular weight of FOXE3 may be higher than the predicted molecular weight as a result of post-translational modifications of the protein.

Rated 4 out of 5 by from Hard to set, but works fineWe bought this product for a western blot on mouse lens epithelium. It took some time to find the right parameters to run the experiment, but when we found them, the antibody worked fine for our samples, and allowed quantification of FOXE3. The only problem remains the aspecific band at 50kDa, it would be interesting to understand what it corresponds to (obviously not FOXE3).
