FcRn Antibody (B-8) is a mouse monoclonal IgG2a kappa light chain antibody that detects FcRn protein of human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). FcRn antibody (B-8) is available in non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. FcRn protein, also known as FCGRT, plays a crucial role in the immune system by mediating immunoglobulin G (IgG) transport across cellular barriers, particularly in the intestinal epithelium of neonatal mice and rats, where FcRn facilitates selective uptake of IgG from maternal milk, providing passive immunity to newborns. This function is vital for neonatal survival, helping newborns quickly acquire antibodies necessary to combat infections before immune system development. FcRn structure consists of a heavy chain and β-2-microglobulin, with the heavy chain showing approximately 35% amino acid identity to major histocompatibility complex (MHC) class I molecules. This structural similarity enables FcRn to effectively bind and recycle IgG, protecting against lysosomal degradation and ensuring IgG remains the longest-lived plasma protein, essential for maintaining long-term immunity.

We have recently performed a Western blot with an anti-FcRn antibody and detected two bands for FcRn for RD lysates, presumably one of which is glycosylated. However, in this image, there seems to be only one band of interest

Does conjugated antibody work for flow cytometry? answer SC provided in past. "The conjugated primary antibody is recommended and warrantied for flow cytometry (FCM), however we are still gathering data to show on our website. Check back for updates!"

Does this antibody block FcRn-mediated internalization of human IgGs in cell lines? If so, what concentration should be used to block the internalization?

Thank you for your question. We do not have any reason to believe that this antibody would block FcRn-mediated internalization. However, we have not tested for this or conducted any research to to support one way or another.

Does this antibody have reactivity to Mouse FcRn(Fcgrt)?? If so, Can I use fluorescence conjugated antibody for flow cytometry??

Is it specific for any particular type of FcRN, like FcgR1, II or III? Or we measure total FcRn with this antibody?

Thank you for your question. This monoclonal antibody is raised against amino acids 24-297 mapping within an N-terminal extracellular domain of FcRn of human origin. The exact epitope recognized by this monoclonal antibody has not been determined. The human gene symbol is FCGRT and the protein accession number for the antigen is P55899.

Is FCRN antibody conjugated with FITC suitable for flow cytometry? If yes, what's the recommended dilution? Thanks.

The conjugated primary antibody is recommended and warrantied for flow cytometry (FCM), however we are still gathering data to show on our website. Check back for updates! Can I use this mouse monoclonal antibody for blocking the FcRn receptor in human cell lines? I pretend block the FcRn receptor during the cell culture (functional studies/inhibition/blocking receptors).

Can I use this mouse monoclonal antibody for immunohistochemical staining of mouse tissue? Or even double or triple staining on the same samples?
