FBXO11 Antibody (E-9) is a mouse monoclonal IgG2b kappa light chain antibody that detects FBXO11 protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). Anti-FBXO11 antibody (E-9) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. FBXO11, also known as F-box only protein 11 or VIT1, is a 927 amino acid nuclear protein that plays a crucial role in the ubiquitin-proteasome pathway, functioning as a substrate recognition component of the SCF (Skp1-CUL-1-F-box protein) E3 ubiquitin ligase complex. This complex is essential for the targeted degradation of specific proteins, including cyclins and p53, which are vital for regulating the cell cycle and maintaining genomic stability. FBXO11′s ability to bind and inhibit p53′s transcriptional activity highlights FBXO11′s importance in cellular responses to stress and tumor suppression. Additionally, reduced FBXO11 expression has been linked to vitiligo, a condition characterized by the loss of skin pigmentation, underscoring FBXO11′s potential role in skin health and disease. Multiple FBXO11 isoforms arise from alternative splicing, which may contribute to FBXO11′s diverse functional roles in various cellular contexts.

Rated 4 out of 5 by from Features of FBXO11(sc-393229)FBXO11 I bought the antibody two months ago, and the results of my IHC experiment using FBXO11 (sc-393229) was relatively better. Here is the detail. 1.Antibody concentrations : 1/250 2.tissue : invasive breast cancer;normal breast tissue 3.Results: 1)high expression of FBXO11 in breast tumor tissue, not expressed in normal tissues ; 2)reduced expression of FBXO11 in carcinoma after Neoadjuvant Chemotherapy. Experimental Procedure 1. Dry frozen tissue sections of 8 µm thickness at room temperature for 2 hours. 2. Fix sections with 4% paraformaldehyde for 15 minutes at room temperature. 3. Wash slides 2X, 4 minutes each, with TBS. 4. Counterstain for 1 minute with hematoxylin. 5. Dehydrate slides with sequential ethanol washes of 1 minute each starting with 75%, followed by 80% 、85%、90%、95%finishing with a 100% ethanol wash. 6. Block endogenous peroxidase by incubating 30 minutes in TBS. 7. Wash slides 3X, 3 minutes each, with TBS. 8. Block non-specific binding sites with 1/250diluted Human anti-mouse antibody for 20 minutes. 9. 1 hour at room temperature, with the appropriate antibody. 10. Wash slides 4 times in TBS, incubate the slides with biotinylated secondary antibody for 30 minutes. 11. Add avidin-biotin-peroxidase reagents, and reveal the resulting peroxidase activity by incubating the slides with a 0.5 mg HRP substrate solution . 12. Wash slides 4X in TBS. 13. Counterstain for 1 minute with hematoxylin. 14. Dehydrate slides with sequential ethanol washes of 1 minute each starting with 75%, followed by 80% 、85%、90%、95%finishing with a 100% ethanol wash. 15. Seal slides. 16. Analyze by optical microscopy.
