FAP Antibody (73.3) is a mouse monoclonal IgG1 antibody that detects fibroblast activation protein (FAP) in mouse and rat samples through applications such as western blotting (WB), immunoprecipitation (IP), and enzyme-linked immunosorbent assay (ELISA). FAP is a crucial cell surface glycoprotein and serine protease primarily expressed in fetal mesenchymal tissues and epithelial cancer fibroblasts, playing a significant role in both cancer progression and embryonic development. In the context of cancer, FAP promotes cellular proliferation, thereby contributing to tumor growth and metastasis, while during embryonic development, FAP is essential for the remodeling of developing tissues, ensuring proper organ formation and function. Structurally, FAP is an integral membrane gelatinase composed of N-glycosylated proteolytically inactive subunits, which are vital for function and regulation. Notably, FAP expression is upregulated on chondrocyte membranes in response to the cytokines interleukin-1 and oncostatin M, and FAP levels have been shown to increase in osteoarthritic patients, indicating potential as a biomarker for disease progression. Furthermore, FAP expression is co-localized with matrix metalloproteinases MMP-1 and MMP-13, as well as CD44 variants, highlighting FAP′s involvement in extracellular matrix remodeling and cellular interactions. Interestingly, studies on mice lacking all copies of the FAP gene have revealed that mice remain fertile but exhibit developmental defects and altered cancer susceptibility, underscoring FAP′s importance in both normal physiology and disease states.
