FANCD2 Antibody (FI17) is a mouse monoclonal IgG1 kappa light chain antibody that detects FANCD2 protein of human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and immunohistochemistry. Anti-FANCD2 antibody (FI17) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. FANCD2 plays a crucial role in the DNA damage response, particularly in the repair of interstrand cross-links, which are a type of DNA damage that can lead to genomic instability and cancer if not properly repaired. FANCD2 is part of the Fanconi anemia (FA) pathway, a critical cellular mechanism that maintains genomic integrity. When DNA is damaged, FANCD2 undergoes monoubiquitination, which is essential for activation and subsequent localization to sites of DNA damage, where FANCD2 interacts with other FA proteins and the breast cancer susceptibility protein BRCA1. This interaction is vital for the recruitment of repair factors to the damaged DNA, thereby facilitating effective repair processes. The human FANCD2 gene is located on chromosome 3p25.3, consists of 44 exons, and encodes a 1,451-amino acid nuclear protein that exists in two isoforms, highlighting its complexity and importance in cellular responses to DNA damage.

Will this antibody give me both the ubiquinated and non-ubiquinated versions of FANCD2. Treatment with cisplatin does not seem to work to give the 2 bands needed (switched cisplatin as well)

Has anyone used this antibody for ChIP-PCR or ChIP-seq? The citations don't seem to be searchable.

How can this antibody be used for double or triple staining, if the other primary antibodies are also raised in mouse?

Thank you for your inquiry. To use more than one primary antibody from the same host in staining, it is recommended to have those antibodies directly conjugated to avoid cross-reaction of secondary antibodies. This FANCD2 antibody is available conjugated to HRP, PE, FITC, AF 488 and AF 647.

Rated 4 out of 5 by from Great AntibodyI use this antibody regularly in different breast cancer cell lines for western blot. It works great ( 1 in 1000, 2%milk-TBST). However, I find some batches to be weaker than others. So I increase the concentration to 1 in 750 to get a strong signal. I don't recommend to use it for immunocytochemistry as I tried as low as 1:50 but the signal is extremely weak.
