SPON1 encodes F-spondin, an extracellular matrix–associated, secreted glycoprotein enriched in the nervous system that modulates cell–matrix interactions, neurite outgrowth, and axon guidance during development and tissue remodeling. F-spondin can influence adhesion and migration by engaging integrin and proteoglycan-dependent signaling and by shaping extracellular matrix organization. It has been linked to regulation of synaptic connectivity and neuronal plasticity, and altered SPON1 expression has been reported in contexts of neurodegeneration and injury-associated remodeling. As an ECM cue, F-spondin is also studied for its roles in vascular and fibrotic processes where matrix signaling and cell motility are central.
F-Spondin CRISPR/Cas9 KO Plasmid (h) is a pool of plasmids designed for targeted disruption of the SPON1 gene in human cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the SPON1 together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the SPON1 open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish F-Spondin protein expression.
This CRISPR knockout system enables efficient generation of SPON1-deficient cell models for investigation of F-Spondin signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
