ETO Antibody (3H11) is a mouse monoclonal IgG2a kappa light chain antibody that detects the ETO protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), and enzyme-linked immunosorbent assay (ELISA). Anti-ETO antibody (3H11) is available as the non-conjugated isotype antibody. ETO, along with its homolog ETO-2 (also known as MTG8 and MTG16, respectively), belongs to the ETO transcription factor family, which is characterized by a zinc-finger domain and four conserved domains essential for their function. Domain II is crucial for dimerization among family members, allowing for complex formation that is vital for their regulatory roles in gene expression. ETO protein is primarily located in the nucleus, where ETO plays a significant role in mediating interactions between DNA-binding proteins and transcriptional regulators, such as the nuclear receptor corepressor (N-CoR). This interaction influences chromatin remodeling and transcriptional repression, which are critical processes in cellular differentiation and development. ETO′s relevance is underscored by involvement in the t(8;21) translocation, leading to the formation of the AML-1/ETO oncoprotein, which is implicated in approximately 12-15% of acute myelogenous leukemia cases of the M2 subtype. The fusion protein disrupts normal differentiation pathways and promotes oncogenic transcriptional repression, highlighting ETO′s importance in both normal cellular function and disease pathology.
