Estrogen Receptor alpha Antibody (D-12) is a mouse monoclonal IgG2a kappa light chain antibody raised against amino acids 2-185 mapping at the N-terminus of human Estrogen Receptor alpha (ERα). ERα monoclonal antibody (D-12) detects ERα of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry on paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). ERα protein is primarily located in the nucleus of estrogen-responsive cells, where ERα functions as a ligand-activated transcription factor belonging to the steroid/thyroid hormone receptor superfamily. Nuclear localization of ERα is crucial because ERα allows direct interaction with DNA at estrogen response elements (EREs), regulating transcription of genes involved in cell proliferation, differentiation, and reproductive functions. This localization is essential for mediating physiological effects of estrogen, including normal sexual development and reproductive health. Mislocalization or dysregulation of ERα can lead to altered gene expression and is associated with various diseases, such as hormone-dependent cancers like breast cancer. ERα monoclonal antibody (D-12) is available in both non-conjugated and multiple conjugated forms, including HRP, FITC, agarose, PE, and various Alexa Fluor® conjugates, providing versatility for a wide range of research applications in studying ERα′s function, localization, and role in disease.
