Estrogen Receptor alpha Antibody (AER314) is a mouse monoclonal IgG1 raised against Estrogen Receptor alpha purified from bovine uterus, detecting human and bovine ERα by western blotting (WB), immunoprecipitation (IP), and immunofluorescence (IF). Estrogen Receptor alpha (ERα) is a ligand-activated transcription factor belonging to the nuclear hormone receptor family and plays a crucial role in regulating gene expression involved in reproductive development and function. Structurally, ERα comprises several domains: the N-terminal A/B domain containing the activation function-1 (AF-1) region responsible for ligand-independent transcriptional activation; the DNA-binding domain (DBD) that allows binding to specific estrogen response elements (EREs) in DNA; the hinge region, which provides flexibility and contains nuclear localization signals; and the C-terminal ligand-binding domain (LBD), which binds estrogen and contains activation function-2 (AF-2) for ligand-dependent transcriptional activation. The structural configuration of ERα enables estrogen binding to induce conformational changes in the LBD, facilitating coactivator protein recruitment and initiating transcription of estrogen-responsive genes. Alterations or mutations in these structural domains can disrupt normal receptor function, leading to aberrant gene expression and are implicated in various diseases, including hormone-dependent cancers like breast cancer. Understanding ERα structure is essential for developing targeted therapeutics and diagnostic tools.
