ESET Antibody (A-7) is a mouse monoclonal IgG1 antibody that detects ERG-associated protein with SET domain (ESET) in mouse, rat, and human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). ESET is a crucial nuclear protein belonging to the histone-lysine methyltransferase family, specifically the Suvar3-9 subfamily, and plays a significant role in regulating chromatin structure. This highly conserved protein, consisting of 150 amino acids, methylates Lys-9 of histone H3, a modification serving as an epigenetic marker for transcriptional repression. Such methylation recruits heterochromatin protein 1 (HP1) to methylated histones, influencing gene expression and maintaining genomic stability. ESET shows predominant expression in testis, with SETDB1 gene located on chromosome 1q21, and mutations can lead to loss of methyltransferase activity, underscoring ESET′s critical role in cellular function and development. Anti-ESET antibody (A-7) serves as an essential tool for researchers studying epigenetic regulation and chromatin dynamics in various biological contexts.

What is the recommended fixation method for immunofluorescence staining with ESET (A-7): sc-271553 monoclonal antibody?

Asked by: AbPolly
Thank you for your inquiry. We recommend fixing cells for 5 minutes in -10°C methanol and allowing to air dry. The full protocol can be viewed here: https://www.scbt.com/scbt/resources/protocols/immunofluorescence-cell-staining
