ERK 2 Antibody (12A4) is a mouse monoclonal IgG1 antibody raised against amino acids 200-250 of human ERK2, a crucial serine/threonine kinase in the MAPK/ERK signaling pathway that orchestrates fundamental cellular processes such as proliferation, differentiation, survival, transcriptional regulation, and development. Extracellular Signal-Regulated Kinase 2 (ERK2), also known as MAPK1 or p42 MAPK, is activated via dual phosphorylation by upstream kinases MEK1 and MEK2, after which ERK2 translocates to the nucleus to phosphorylate a myriad of substrates including transcription factors like ELK1, c-Fos, c-Myc, and Ets1/2, as well as cytoskeletal proteins and other kinases such as RSK and MSK. ERK2 interacts with scaffold proteins like KSR and MP1, and is regulated by phosphatases such as MKP-1 and MKP-3. Aberrant ERK2 signaling is associated with various pathologies including cancer, making ERK2 a significant target in biomedical research. ERK2 monoclonal antibody (12A4) exhibits species reactivity with mouse, rat, and human samples, and is recommended for use in western blotting (WB) and immunoprecipitation (IP) applications, providing a valuable tool for researchers studying signal transduction pathways, protein-protein interactions, cellular responses to growth factors and stress stimuli, and the molecular mechanisms underlying disease states.
