ERK 1/2 Antibody (MK1) is a mouse monoclonal IgG1 kappa light chain antibody raised against amino acids 325-345 of ERK1 of rat origin, specifically detecting both ERK1 (p44 MAPK) and ERK2 (p42 MAPK) proteins in mouse, rat, canine, and human samples by western blotting (WB), immunoprecipitation (IP), and immunofluorescence (IF). ERK1 and ERK2 are closely related serine/threonine kinases that play a pivotal role in the mitogen-activated protein kinase (MAPK)/ERK signaling pathway, a critical conduit for transducing extracellular signals such as growth factors, cytokines, hormones, and environmental stresses into intracellular responses, thereby regulating fundamental cellular processes including proliferation, differentiation, survival, migration, and gene expression. Activation of ERK1/2 involves phosphorylation by upstream kinases MEK1 and MEK2, which are themselves activated by Raf kinases following stimulation of receptor tyrosine kinases (RTKs) and G protein-coupled receptors (GPCRs). Phosphorylated ERK1/2 translocate to the nucleus where they phosphorylate a variety of transcription factors such as ELK1, c-Fos, c-Myc, and CREB, influencing gene expression and driving cell cycle progression and other essential functions. ERK1/2 also phosphorylate cytoplasmic targets, affecting cytoskeletal rearrangements and cell motility; dysregulation of ERK1/2 signaling is implicated in numerous pathological conditions, notably various cancers, neurodegenerative diseases, and developmental disorders, making ERK1/2 crucial targets in biomedical research and therapeutic development.
