ErbB-3 Antibody (G-4) is a mouse monoclonal IgG2b kappa light chain antibody raised against amino acids 1315-1342 at the C-terminus of human ErbB-3, a crucial member of the Erb-B receptor tyrosine kinase family. ErbB-3 mouse monoclonal antibody (G-4) recognizes an epitope within this C-terminal region and demonstrates broad species cross-reactivity, detecting ErbB-3 in mouse, rat, human, bovine, and porcine samples through Western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), flow cytometry (FCM), and enzyme-linked immunosorbent assay (ELISA) applications. Anti-ErbB-3 monoclonal antibody (G-4) is available in non-conjugated form and multiple conjugated forms including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and various Alexa Fluor® conjugates, offering versatility for diverse experimental needs. ErbB-3, also known as FERLK, HER3, LCCS2, MDA-BF-1, VSCN1, erbB3-S, and ERBB3, functions as a sophisticated signal transducer in cellular pathways controlling growth, survival, and differentiation. ErbB-3′s signaling mechanism involves formation of either homodimers or heterodimers with other family members, particularly Neu (ErbB-2/HER2), following binding of ErbB-3′s primary ligand, heregulin. This dimerization triggers activation of downstream pathways, notably the phosphoinositide 3-kinase (PI3K) cascade, through phosphorylation of specific YXXM motifs in ErbB-3′s C-terminal region. The clinical significance of ErbB-3 becomes evident in cancer biology, where ErbB-3 overexpression frequently occurs in various carcinomas, including human mammary tumors. This dysregulation contributes to both tumor progression and resistance to therapeutic interventions by enhancing oncogenic signaling and cell survival mechanisms. Through these diverse cellular roles, ErbB-3 (G-4) antibody serves as an invaluable tool for investigating cancer development mechanisms and therapeutic resistance pathways.

sc-7390 replaced product sc-285. Was the same epitope used for sc-7390 as used for the sc-285 antibody? I am trying to find information regarding epitope mapping for the discontinued sc-285 antibody (which mapped to 10 D3).

Is this erbb3 antibody should be applied as intercellular staining ? or as surface staining

Thank you for your question. ErbB-3 Antibody (G-4): sc-7390 is specific against amino acids 1315-1342. The C-terminal amino acid range between 665 – 1342 is cytoplasmic, so the antibody would recognize only the cytoplasmic part of its target protein.

How should the blocking peptide sc-7390 P be used?

Antigen retrieval was performed in citrate buffer, 30'. Primary antibody: 1:50 dilution, 1h, RT. Secondary antibody: Alexa Fluor 488 (1:500), 1h, RT. Results: negative; nonspecific signal was observed both in epithelial and connective fractions. Immunocytochemistry: the antibody was tested on porcine mammary epithelial cells isolated from 6 months old gilts. Cells were fixed with acetone-methanol (1:1) and stained as above (see immunohistochemistry). Primary antibody was diluted 1:100. Result: negative.

Rated 1 out of 5 by from Did not work for IPI requested a sample of this antibody to IP HER3 from human colorectal cancer cells with 1ug antibody per 200ug of proteins from cell lysate. HER3 in these cells were able to be IP pulled down or detected by Western blotting with a HER3 antibody carried by Cell Signaling.
