ENC1 (ectodermal-neural cortex 1) encodes an actin-binding protein of the kelch repeat family that links cytoskeletal remodeling to cellular differentiation programs. In human cells, ENC1 has been associated with regulation of actin dynamics, oxidative stress responses, and context-dependent control of proliferation and neuronal lineage features, supporting roles in development and cell state maintenance. Altered ENC1 expression has been reported across multiple tumor types and in models of neurobiology, making it a useful node for studying pathways that couple redox signaling, cytoskeletal organization, and transcriptional outputs. ENC1 CRISPR/Cas9 KO Plasmid (h) is a pool of plasmids designed for targeted disruption of the ENC1 gene in human cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the ENC1 together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the ENC1 open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish ENC1 protein expression.
