Elastin Antibody (BA-4) is a mouse monoclonal IgG1 kappa light chain antibody that detects elastin protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and immunohistochemistry. Anti-elastin antibody (BA-4) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Elastin plays a crucial role in providing elasticity and resilience to connective tissues, particularly in the extracellular matrix of arteries, where elastin is essential for maintaining vascular integrity and function. Elastin′s unique structure, characterized by a high content of glycine, proline, and hydrophobic residues, allows elastin to form a rubber-like network through lysine-derived crosslinks, such as desmosines. This structural property is vital for the ability of tissues to stretch and recoil, which is particularly important in organs that undergo significant mechanical stress, such as the lungs and blood vessels. As individuals age, the cross-linking of elastin with collagen can lead to a decrease in tissue elasticity, contributing to various age-related disorders. Furthermore, deficiencies in elastin are linked to serious conditions like supravalvular aortic stenosis and Williams-Beuren syndrome, highlighting elastin′s importance in cardiovascular health. The human elastin gene is located on chromosome 7q11.23, underscoring elastin′s genetic significance in developmental and pathological contexts.

We used this antibody (1:500) for a western blot. We could see bands at about 30 kDa and 25 kDa and weak bands at 20 and 15 kDa. We could see that other research groups have the same problem. Are there any ideas why?

For western blotting, this antibody gives a strong band ~30kDa (expected size is 60-70kDa) in human smooth muscle cell lysates. Do you recommend to use this antibody in non-reducing conditions and perhaps PAGE gel?

We want to use this antigen for elastin detection in mouse (formalin-fixed, paraffin-embedded tissue sections). Do I need to epitope retrieval ? In that case, Would you please tell me optimum conditions for epitope retrieval.

Thank you for your question. We do recommend performing antigen retrieval when using this antibody on formalin-fixed paraffin-embedded tissue sections. We recommend using heat treatment with 10 mM sodium citrate buffer (pH 6.0). Does this mouse monoclonal work for IHC or IF staining of mouse tissue?

Yes. We recommend using a directly conjugated mouse monoclonal primary antibody. This mouse monoclonal antibody for elastin is available conjugated to HRP (sc-58756 HRP), 200 µg/ml, for WB, IHC(P) and ELISA; and to either phycoerythrin (sc-58756 PE) or fluoroscein (sc-58756 FITC), Alexa Fluor® 488 (sc-58756 AF488) or Alexa Fluor® 647 (sc-58756 AF647), 200 µg/ml, for IF, IHC(P) and FCM.

Fairly faint result despite doubling protein and using a 1:200 dilution as recommended. Antibody should fall between ladder bands 3 and 4 (98 and 64 kDa) according to reported MW of 71 kDa. Strongest band shows around ~50 kDa, but faint bands also present around that range.
