eIF5A Antibody (H-8) is a mouse monoclonal IgG2a kappa light chain antibody that detects eIF5A protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). anti-eIF5A antibody (H-8) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. eIF5A plays a crucial role in cellular processes, particularly in translation initiation, where eIF5A is essential for proper functioning of ribosomal peptidyl-transferase. eIF5A is encoded by the human eukaryotic translation initiation factor 5A gene, also known as eIF-4D or eIF5A1, which is located on chromosome 17p13 and produces a 154 amino acid protein vital for maintaining cellular polyamine homeostasis. eIF5A1 is localized in both nuclear and cytoplasmic compartments of mammalian cells, where eIF5A1 not only stimulates translation but also may facilitate nucleocytoplasmic mRNA transport. A unique aspect of eIF5A1 is post-translational modification at Lys-50, where hypusine is formed, a modification critical for biochemical activity and necessary for cellular proliferative signaling. Furthermore, eIF5A1 serves as a cofactor for Rev transactivator protein of human immunodeficiency virus type 1 (HIV-1), and disruption of eIF5A1 interaction with Rev can inhibit viral replication cycle. Additionally, eIF5A2, a highly conserved protein with 94% sequence similarity to eIF5A1, is implicated in cancer biology, as eIF5A2 gene amplification is observed in ovarian carcinomas, linking eIF5A2 overexpression to advanced stages of ovarian cancer.
