EDEM2 Antibody (JD-32) is a mouse monoclonal IgG1 kappa light chain antibody that detects EDEM2 protein of human origin by immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). Anti-EDEM2 antibody (JD-32) is available as a non-conjugated format. EDEM2, or ER degradation-enhancing alpha-mannosidase-like 2, plays a crucial role in the endoplasmic reticulum (ER) quality control system, specifically in the process known as ER-associated degradation (ERAD). This pathway is essential for maintaining cellular homeostasis by ensuring that misfolded or improperly assembled proteins are efficiently identified and targeted for degradation. EDEM2 is a 578 amino acid secreted protein that works in concert with other EDEM family members, such as EDEM and EDEM3, to facilitate the retrotranslocation of terminally misfolded proteins from the ER to the cytoplasm, where these proteins are subsequently degraded by proteasomes. EDEM2 contains a mannosidase homology domain, an N-terminal cleavable signal sequence, and a C-terminal extension that is vital for ER retention and overall functionality. Human EDEM2 shares 93% sequence identity with its mouse counterpart, indicating a conserved role across species, and two EDEM2 isoforms arise from alternative splicing events, further highlighting the complexity and regulatory mechanisms governing protein quality control in the ER.

Can sc-130460: EDEM2 (JD-32) monoclonal antibody be used to stain formalin-fixed, paraffin-embedded (FFPE) tissue sections?

Asked by: cjMara
Thank you for your question. Yes, sc-130460: EDEM2 (JD-32) is recommended for use in IHC with paraffin-embedded sections. We recommend performing antigen retrieval with sodium citrate buffer (pH 6) and heat. The full protocol can be found here: https://www.scbt.com/scbt/resources/protocols/immunoperoxidase-staining
