E6-AP Antibody (E-4) is a mouse monoclonal IgG2b kappa light chain antibody that detects E6-AP of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). anti-E6-AP antibody (E-4) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. E6-AP, also known as ubiquitin protein ligase E3A (UBE3A), plays a crucial role in the ubiquitin-proteasome pathway, which is essential for regulating protein degradation and maintaining cellular homeostasis. UBE3A is primarily located in the nucleus and cytoplasm, where UBE3A facilitates the ubiquitination of target proteins, thereby marking them for degradation by the 26S proteasome. Proper localization of UBE3A is vital, as UBE3A ensures that substrates such as the tumor suppressor protein p53 are effectively regulated; dysregulation can lead to the accumulation of p53 in the cytoplasm, contributing to oncogenic processes and disorders like Angelman syndrome. UBE3A′s interaction with the human papillomavirus E6 oncogene further underscores UBE3A′s importance in cancer biology, as this interaction enhances the ubiquitination of p53, promoting its degradation and allowing for unchecked cellular proliferation. Genetic mutations that impair UBE3A function are linked to Angelman syndrome, a neurodevelopmental disorder characterized by severe motor dysfunction and cognitive impairment, highlighting UBE3A′s critical role in both cancer and neurodevelopmental health.

Can E6-AP (E-4): sc-166689 monoclonal antibody be used to stain formalin-fixed, paraffin-embedded (FFPE) tissue sections?

Asked by: jerojero
Thank you for your question. Yes, E6-AP (E-4): sc-166689 has been validated for use in IHC with paraffin-embedded sections. We recommend performing antigen retrieval with sodium citrate buffer (pH 6) and heat. The full protocol can be found here: https://www.scbt.com/scbt/resources/protocols/immunoperoxidase-staining
