NFIL3 encodes the basic leucine zipper transcription factor E4BP4, a context-dependent regulator of gene expression that integrates circadian cues with immune and metabolic programs. E4BP4 influences differentiation and effector function across lymphoid lineages, including regulation of cytokine networks and survival pathways, and contributes to homeostatic control of inflammatory signaling. It also interfaces with transcriptional circuits that shape cellular stress responses and developmental timing, linking rhythmic regulation to broader gene regulatory networks. Dysregulated NFIL3/E4BP4 activity has been associated with immune-mediated pathology, hematologic phenotypes, and altered metabolic and circadian phenotypes in research settings.
E4BP4 CRISPR/Cas9 KO Plasmid (h) is a pool of plasmids designed for targeted disruption of the NFIL3 gene in human cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the NFIL3 together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the NFIL3 open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish E4BP4 protein expression.
