DNA-PKCS Antibody (G-12) is a mouse monoclonal IgG2b kappa light chain antibody that detects DNA-PKcs in mouse, rat, and human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). anti-DNA-PKcs antibody (G-12) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. DNA-PKcs plays a crucial role in cellular response to DNA double-strand breaks (DSBs), which are among the most lethal forms of DNA damage. DNA-PKcs is primarily located in the nucleus, where DNA-PKcs is essential for the non-homologous end joining (NHEJ) repair pathway, a critical mechanism for maintaining genomic stability. Deficiencies in DNA-PKcs can lead to severe consequences, including increased susceptibility to cancer and other genetic disorders, as the inability to repair DSBs can result in chromosomal aberrations and cell death. DNA-PKcs is part of a larger complex that includes the Ku proteins, which are vital for recognizing and binding to DSBs, thereby facilitating the repair process. The significance of DNA-PKcs in the NHEJ pathway underscores DNA-PKcs potential as a therapeutic target in cancer treatment, where enhancing or inhibiting DNA-PKcs activity could influence tumor response to radiation and chemotherapy.

The use of mouse monoclonal antibodies with mouse samples is very common and typically poses no problem. To eliminate any potential cross-reactivity of an anti-mouse secondary detection reagent, our DNA-PKCS sc-390849 is available in a variety of direct conjugations, such as AC, HRP, PE, FITC, AlexaFluor 488 and AlexaFluor 647.

Rated 3 out of 5 by from This antibody can detect DNA-PKcs by Western blotWe acquired this antibody approximately 7 months ago. We performed a western blot analysis of HeLa cells whole cell lysate using a 1 to 1000 dilution. We used four different samples of untreated HeLa cells and got different levels of signals using the same concentration of protein. Although the antibody works, it was not consistent.
