DNA Ligase IV Antibody (D-8) is a mouse monoclonal IgG2a kappa light chain antibody that detects DNA Ligase IV of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). DNA Ligase IV (D-8) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. DNA Ligase IV plays a crucial role in repairing double-strand breaks in DNA, which is vital for maintaining genomic stability and preventing mutations that can lead to cancer. DNA Ligase IV operates with X-ray repair cross-complementing protein XRCC4, forming a complex that enhances DNA repair efficiency by preferentially binding to DNA with nicks or broken ends. This interaction is particularly important during V(D)J recombination, a process essential for adaptive immune system development, where double-strand breaks are generated to facilitate immunoglobulin and T-cell receptor gene rearrangement. Disruptions in DNA Ligase IV or XRCC4 function can lead to severe consequences, including increased apoptosis in developing neurons and impaired maturation of B and T cells, highlighting DNA Ligase IV′s importance in both DNA repair and immune system development.

Can DNA Ligase IV (D-8): sc-271299 monoclonal antibody be used in IF or IHC with mouse tissue or cells? Will there be non-specific staining?

Thank you for your inquiry. The use of mouse monoclonal antibodies with mouse samples is very common and typically poses no problem. To eliminate any potential cross-reactivity of an anti-mouse secondary detection reagent, DNA Ligase IV (D-8): sc-271299 is available in a variety of direct conjugations, such as HRP, PE, FITC and AlexaFluors.

Rated 4 out of 5 by from The antibody works.We acquired this antibody 7 months ago and probed on whole cell lysates of HeLa cells. We used a 1 to 1000 dilution with a 3 minute exposure. We saw a decent band. It works.

We see no detection of endogenous protein in U2OS and primary human fibroblasts, in fact it detects multiple bands, none of which are specific as adjudged by transfection with siRNA against DNA ligase IV.
