Dkk-1 Antibody (B-7) is a mouse monoclonal IgG1 kappa light chain antibody that detects Dkk-1 of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). Dkk-1 (B-7) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Dkk-1, also known as dickkopf WNT signaling pathway inhibitor 1, plays a crucial role in regulating Wnt signaling, which is essential for numerous developmental processes such as embryogenesis and central nervous system patterning. Specifically, Dkk-1 functions as an antagonist to Wnt signaling by binding to the frizzled receptor, thereby preventing the activation of dishevelled and the subsequent accumulation of β-catenin. This inhibition is vital for maintaining proper cellular differentiation and tissue morphogenesis. Dkk-1 is a 266-amino acid secreted protein characterized by a 31-residue N-terminal signal peptide, two cysteine-rich domains, and a potential carboxy-terminal N-glycosylation site. Human Dkk-1 transcripts are predominantly expressed in fetal kidney, adult placenta, and adult prostate, indicating importance in both developmental and reproductive contexts. Regulatory elements upstream of the Dkk-1 gene suggest a complex interplay of transcription factors, including p53, Sp1, and GATA family members, which may influence expression in various tissues.
