Dia 1 Antibody (E-4) is a mouse monoclonal IgG1 kappa light chain antibody that detects Dia 1 protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). Dia 1 (E-4) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Dia 1 protein, also known as DIAPH1 (diaphanous homolog 1) or DRF1, plays a crucial role in regulating actin dynamics, which is essential for various cellular processes such as cell shape, motility, and division. DIAPH1 is characterized by unique structure, including tandemly aligned formin homology 1 (FH1) and formin homology 2 (FH2) domains that facilitate nucleation and elongation of actin filaments. DIAPH1 contains a diaphanous autoregulatory domain (DAD) that modulates activity through autoinhibition, as well as a Rho GTPase-binding/formin homology 3 (GBD/FH3) domain that interacts with DAD domain. DIAPH1 localization to cell membrane allows direct regulation of actin polymerization at sites of cellular activity. Mutations in DIAPH1 gene have been associated with deafness autosomal dominant type 1 (DFNA1), highlighting DIAPH1′s importance in auditory function.
