SEE ALSO...

DFNA5 Antibody (G-9) is a mouse monoclonal IgG2a kappa light chain antibody that detects DFNA5 protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). anti-DFNA5 antibody (G-9) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. DFNA5, also known as ICERE-1, consists of 496 amino acids and is primarily expressed in cochlear tissue, as well as in the placenta, brain, heart, liver, lung, and pancreas, where DFNA5 exists in two alternatively spliced isoforms, referred to as short and long. DFNA5 plays a crucial role in auditory function; mutations in the gene encoding DFNA5 are associated with non-syndromic sensorineural deafness autosomal dominant type 5, a condition that leads to hearing loss due to damage in the auditory pathways of the brain. DFNA5 is located on human chromosome 7, which contains over 1,000 genes and accounts for nearly 5% of the human genome. Notably, defects in other genes on chromosome 7 have been implicated in various genetic disorders, including Osteogenesis imperfecta and Williams-Beuren syndrome, highlighting the critical nature of this chromosomal region in human health.

I am using DFNA5 (G-9): sc-393162 monoclonal antibody to stain paraffin embedded tissue. Do you recommend antigen retrieval when using this antibody?

Asked by: Germaine
We recommend performing antigen retrieval with sodium citrate buffer (pH 6) and heat. The full protocol can be found here: https://www.scbt.com/scbt/resources/protocols/immunoperoxidase-staining

Rated 4 out of 5 by from Not work that wellthis antibody not work that well on our mouse tissue, maybe because the size is too close to the IgG heavy band,
Date published: 2024-10-16
Rated 1 out of 5 by from Does not work on WB with endogenous expressionThis antibody does not work on Western blot with human cell line samples that endogenously express DFNA5/GSDME (or does so too weakly to definitively ascertain). Positive (NCI-H522) and negative (HCT-15) control samples that have been validated with several other commercial antibodies were used.

Rated 4 out of 5 by from Great for WBGreat product for detecting exogenous DFNA5 expression in Hela cells and HaCaT cells. However, we are not sure that whether this antibody is able to detect endogenous DFNA5 expression(we detected two bands at about 50KD in both Hela WT and Hela DFNA5-KO cells). We also did not find the band of cleaved DFNA5 when we tried to activate DFNA5.
