DEF-3 Antibody (D-3) is a mouse monoclonal IgG2b kappa light chain antibody that detects DEF-3 protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). Anti-DEF-3 antibody (D-3) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. DEF-3 plays a crucial role in RNA binding and is part of an evolutionarily conserved family of RNA binding proteins, which also includes LUCA15. Both DEF-3 and LUCA15 are predominantly expressed in the adult heart and thymus, as well as in fetal kidney tissue, while expression is significantly lower in the fetal thymus and adult kidney. Notably, in the hematopoietic system of mice, DEF-3 expression is downregulated during the differentiation of progenitor cells into granulocytes, yet remains present during macrophage development. Structural features of DEF-3 include two zinc finger motifs, a bipartite nuclear localization signal, and two RNA binding motifs, which are essential for binding poly(G) RNA. Human DEF-3 and LUCA15 genes are located on chromosome 3p21.3, a region often deleted in small cell lung cancer cell lines, highlighting potential implications in cancer biology, particularly relating to mutations found in dominant and recessive oncogenes within this chromosomal region.
