DD1-4 Antibody (H-8) is a mouse monoclonal IgG2a antibody that detects dihydrodiol dehydrogenase (DD1, DD2, DD3, and DD4) in human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). Anti-dihydrodiol dehydrogenase antibody (H-8) recognizes multiple isoforms of dihydrodiol dehydrogenase, which are part of the aldo-oxo reductase/aldo-keto reductase superfamily and play a crucial role in the metabolism of xenobiotics, including polycyclic aromatic hydrocarbons, pesticides, and steroid hormones. These enzymes utilize NADH or NADPH as cofactors to catalyze the reduction of ketone-containing drugs, with DD1 and DD2 specifically functioning as 20α-hydroxysteroid dehydrogenases that convert progesterone into its inactive form, 20α-hydroxyprogesterone. Notably, DD3 and DD4 serve as 3α-hydroxysteroid dehydrogenases, with DD3 uniquely capable of catalyzing the dehydrogenation of trans-benzenedihydrodiol and trans-naphthalenedihydrodiol. The expression patterns of these isoforms vary, as DD1 and DD2 are ubiquitously expressed, while DD4 mRNA is primarily found in the liver, highlighting the tissue-specific roles of these enzymes in steroid metabolism and detoxification processes. Anti-dihydrodiol dehydrogenase antibody (H-8) targeting of these isoforms makes DD1-4 an invaluable tool for researchers studying the biochemical pathways involving dihydrodiol dehydrogenases and their implications in human health and disease.
