DCIR Antibody (G-9) is a mouse monoclonal IgG2b kappa light chain antibody that detects DCIR protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). DCIR (G-9) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. DCIR, or dendritic cell immunoreceptor, is a type II membrane glycoprotein characterized by a single carbohydrate recognition domain (CRD) and is closely related to macrophage lectin and hepatic asialoglycoprotein receptors. DCIR′s intracellular domain features a consensus immunoreceptor tyrosine-based inhibitory motif (ITIM), which plays a crucial role in modulating cellular responses, such as protein tyrosine phosphorylation and B-cell receptor-mediated calcium mobilization. The presence of the phosphorylated ITIM motif allows for the binding of SH2 domain-containing phosphatases, thereby influencing various signaling pathways. DCIR is predominantly expressed on dendritic cells, monocytes, macrophages, B lymphocytes, and granulocytes, but is notably absent in natural killer and T cells. Importantly, DCIR expression on dendritic cells is downregulated in response to maturation signals, such as lipopolysaccharides, tumor necrosis factor-alpha, or CD40 stimulation, highlighting its role in immune regulation and the potential impact on dendritic cell function during immune responses.

What blocking solution should I use with DCIR (G-9): sc-374583 monoclonal antibody for Western blot?

Asked by: Germaine
Thank you for your question. We recommend blocking with Blotto (either TBS Blotto A: sc-2333 or TBS Blotto B: sc-2335). Our Western blot protocol can be found here: https://www.scbt.com/scbt/resources/protocols/western-immuno-blotting
