DARPP-32 Antibody (H-3) is a mouse monoclonal IgG1 kappa light chain antibody that detects DARPP-32 protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). DARPP-32 (H-3) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. DARPP-32 protein, also known as dopamine and adenosine 3′,5′-monophosphate-regulated phosphoprotein, plays a crucial role in dopaminergic signaling pathways, which are vital for numerous brain functions, including motor control, reward processing, and cognitive functions. Located predominantly in neurons that receive dopaminergic input, DARPP-32 acts as a key integrator of dopamine signaling, where phosphorylation state is critical for modulating the activity of protein phosphatase 1, a major enzyme involved in synaptic plasticity and memory formation. Phosphorylation of DARPP-32 on threonine is stimulated by dopamine binding to D1-like receptors, while D2-like receptor activation leads to dephosphorylation, highlighting the intricate balance of signaling pathways that govern neuronal function. Abnormalities in DARPP-32 signaling have been implicated in various neurological disorders, including schizophrenia, Parkinson′s disease, and substance abuse, making DARPP-32 an important target for research and therapeutic interventions.

Can This antibody reacts with full-length DARPP-32 and truncated DARPP-32?? or reacts both of them? Thank you.

Thank you for your question. This antibody reacts with full length DARPP-32. According to Uniprot (see link below), the truncated DARPP-32 isoform (also known as t-DARPP) is missing amino acids 1-36. This monoclonal antibody is raised against amino acids 2-34 at the N-terminus of DARPP-32 of human origin, so it would not be expected to detect the truncated isoform.

What is the recommended antigen retrieval method and incubation period for this antibody for IHC (mouse paraffin embedded sections)?

Thank you for your question. We recommend using sodium citrate buffer (pH 6.0) plus heat treatment for antigen retrieval with this antibody. We recommend incubating with the primary antibody for 30 minutes at room temperature, or overnight at 4 degrees C. Application: Immunofluorescence (Frozen mouse brain sections); Fixative: 4% Paraformaldehyde; Permeabilization 0.3% Triton X-100 in PBS; Incubation time overnight incubation at 4C; Dilution 1:200.
