DARPP-32 Antibody (G-5) is a mouse monoclonal IgG2a antibody that detects DARPP-32 in mouse, rat, and human samples through various applications including western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). DARPP-32, or dopamine and adenosine 3′,5′-monophosphate-regulated phosphoprotein, plays a crucial role in dopaminergic signaling pathways, which are vital for numerous brain functions and are often disrupted in neurological disorders such as schizophrenia, Parkinson′s disease, and substance abuse. DARPP-32 is predominantly located in the cytoplasm of neurons that receive dopaminergic input, where DARPP-32 acts as a key regulator of dopamine signaling. The spatial distribution of DARPP-32 is significant because DARPP-32 allows for the integration of signals from both D1-like and D2-like dopamine receptors, facilitating a balanced response to dopamine. Upon activation of D1-like receptors, DARPP-32 is phosphorylated by protein kinase A, which enhances DARPP-32′s ability to inhibit protein phosphatase 1, thereby modulating downstream signaling pathways. Conversely, when D2-like receptors are activated, the inhibition of protein kinase A leads to the dephosphorylation of DARPP-32, demonstrating DARPP-32′s role as a molecular switch in dopaminergic signaling. DARPP-32′s ability to respond to various neurotransmitters further underscores DARPP-32′s importance in maintaining neuronal function and potential as a therapeutic target in treating dopamine-related disorders.
