Datasheets
  • DARPP-32 Antibody (G-5) is a mouse monoclonal IgG2a κ, cited in 2 publications, provided at 200 µg/ml
  • raised against amino acids 134-195 mapping epitope corresponding to amino acids 134-195 of DARPP-32 of bovine origin
  • recommended for detection of DARPP-32 of mouse, rat and human origin by WB, IP, IF, IHC(P) and ELISA; also reactive with additional species, including bovine
  • See DARPP-32 (H-3): sc-271111 for DARPP-32 antibody conjugates, including AC, HRP, FITC, PE, Alexa Fluor® 488, 594, 647, 680 and 790.
  • m-IgG Fc BP-HRP and m-IgGκ BP-HRP are the preferred secondary detection reagents for DARPP-32 Antibody (G-5) for WB and IHC(P) applications. These reagents are now offered in bundles with DARPP-32 Antibody (G-5) (see ordering information below).

DARPP-32 Antibody (G-5) is a mouse monoclonal IgG2a antibody that detects DARPP-32 in mouse, rat, and human samples through various applications including western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). DARPP-32, or dopamine and adenosine 3′,5′-monophosphate-regulated phosphoprotein, plays a crucial role in dopaminergic signaling pathways, which are vital for numerous brain functions and are often disrupted in neurological disorders such as schizophrenia, Parkinson′s disease, and substance abuse. DARPP-32 is predominantly located in the cytoplasm of neurons that receive dopaminergic input, where DARPP-32 acts as a key regulator of dopamine signaling. The spatial distribution of DARPP-32 is significant because DARPP-32 allows for the integration of signals from both D1-like and D2-like dopamine receptors, facilitating a balanced response to dopamine. Upon activation of D1-like receptors, DARPP-32 is phosphorylated by protein kinase A, which enhances DARPP-32′s ability to inhibit protein phosphatase 1, thereby modulating downstream signaling pathways. Conversely, when D2-like receptors are activated, the inhibition of protein kinase A leads to the dephosphorylation of DARPP-32, demonstrating DARPP-32′s role as a molecular switch in dopaminergic signaling. DARPP-32′s ability to respond to various neurotransmitters further underscores DARPP-32′s importance in maintaining neuronal function and potential as a therapeutic target in treating dopamine-related disorders.

Can Darpp-32 (G-5) Ab be used to detect total Darpp-32 from? ( Truncated, Full-length Darpp-32) Additionally, What's the difference between G-5 and F-3 Darpp-32 Ab?

Asked by: bami
Thank you for your question. I have checked this product. The immunogen information of this product is 'raised against amino acids 134-195 mapping epitope corresponding to amino acids 134-195 of DARPP-32 of bovine origin'. Also we do not have the Darpp-32 (F-3) antibody, so I have no idea about the difference between G-5 and F-3. I don't know the F-3 is a monoclonal antibody or a polyclonal antibody and their host species. Usually, in parentheses, such as (G-5) and (F-3) are the clone numbers, which are randomly coded.

Can DARPP-32 (G-5): sc-398360 monoclonal antibody be used to stain formalin-fixed, paraffin-embedded (FFPE) tissue sections?

Asked by: jenniferc
Thank you for your question. Yes, DARPP-32 (G-5): sc-398360 has been validated for use in IHC with paraffin-embedded sections. We recommend performing antigen retrieval with sodium citrate buffer (pH 6) and heat. The full protocol can be found here: https://www.scbt.com/scbt/resources/protocols/immunoperoxidase-staining
