DAP12 Antibody (A-4) is a mouse monoclonal IgG2a kappa light chain antibody that detects DAP12 protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). DAP12 (A-4) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. DAP12, also known as TYROBP, is a crucial phosphoprotein that plays a significant role in the activation of natural killer (NK) cells, which are essential components of the innate immune system. Located in the plasma membrane, DAP12 interacts with several receptor families, including killer-cell inhibitory receptors (KIR), CD94/NKG2, and Ly-49, facilitating the recognition of major histocompatibility complex (MHC) class I molecules. This interaction is vital for the regulation of NK cell activity, as DAP12 helps to balance stimulatory and inhibitory signals, ultimately influencing immune responses. Notably, DAP12′s phosphorylation allows binding to signaling proteins such as ZAP-70 and Syk, indicating that DAP12′s activation pathway may share similarities with those of T and B cell antigen receptors. DAP12′s ability to form complexes with other proteins, such as CD94/NKG2C, Ly-49D, and Ly-49H, further underscores DAP12′s importance in immune signaling and cellular activation, making anti-DAP12 antibody (A-4) an invaluable tool for researchers studying NK cell biology and immune responses.

Has anyone used this antibody in paraffin sections? Thanks.

Thank you for your question. We have not yet tested this mouse monoclonal antibody, DAP12 (A-4): sc-166084, in formalin-fixed paraffin-embedded tissue sections. As an alternative, we do offer a different clone, DAP12 (G-5): sc-133174, which is recommended for IHC(P). However, this antibody has only been tested in human tissues, and reactivity with other species has not yet been determined.
