DAAO Antibody (B-3) is a mouse monoclonal IgG2a kappa light chain antibody that detects DAAO protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). DAAO Antibody (B-3) is available in non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. D-amino acid oxidase (DAAO) plays a crucial role in metabolizing exogenous D-amino acids, which can accumulate during aging, and is essential for modulating D-serine levels in the brain, acting as a detoxifying agent. DAAO specifically targets D-configured amino acids and shows preference for those with small, hydrophobic side chains, as well as polar, aromatic, and basic functional groups. Single base-pair substitutions at specific amino acid residues can lead to DAAO activity loss in mice, highlighting DAAO′s sensitivity to genetic variations. A mutation at residue Y238, a conserved residue in the active site, significantly slows product release compared to wild-type enzyme, underscoring Y238′s importance in regulating active site access. While other conserved residues in the active site don′t participate in acid-base catalysis, these residues remain critical for substrate interactions, suggesting a hybrid transfer mechanism for catalysis influenced by substrate orientation within the active site.
