Cytokeratin 8 Antibody (C51) is a mouse monoclonal IgG1 kappa light chain antibody that detects Cytokeratin 8 in mouse, rat, and human samples through various applications including western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). Anti-Cytokeratin 8 antibody (C51) is available in both non-conjugated and multiple conjugated forms, such as agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and various Alexa Fluor® conjugates. Cytokeratin 8, part of a diverse group of intermediate filament proteins, plays a crucial role in maintaining structural integrity of epithelial cells and is essential for cellular differentiation and tissue specialization. Cytokeratin 8 is predominantly located in cytoplasm of simple epithelial cells, where Cytokeratin 8 forms a network providing mechanical support and resilience against stress. Proper localization of Cytokeratin 8 is vital, as misexpression can lead to pathological conditions, including various carcinomas. Expression of Cytokeratin 8 is often upregulated in epithelial-derived tumors, making Cytokeratin 8 a significant marker for cancer diagnosis and prognosis. Presence of Cytokeratin 8 in squamous cell carcinomas derived from stratified epithelia has been associated with poor clinical outcomes, highlighting potential as a therapeutic target. Cytokeratin 8 and 18 pair is typically expressed in simple epithelia, and abnormal expression in stratified epithelial cells can indicate malignancy, further underscoring importance in cancer research and treatment strategies.

What is the proper antigen retrieval condition suitable for this antibody when staining on FFPE-section?

Asked by: Anonymous
Thank you for your question. You can find our standard protocol for IHC(P) applications by following this link: https://www.scbt.com/resources/protocols/immunoperoxidase-staining The recommended antigen retrieval method is heat treatment, which in this protocol is listed under point "C. Formalin-Fixed, Paraffin-Embedded Tissue Sections"

Can I use this mouse monoclonal antibody for immunohistochemical staining of mouse tissue? Or even double or triple staining on the same samples?

Asked by: DefinitelyNotMatt
Thank you for your question. Yes. We recommend using a directly conjugated mouse monoclonal primary antibody. This antibody is currently available conjugated to phycoerythrin (sc-8020 PE), fluorescein (sc-8020 FITC), Alexa Fluor® 488 (sc-8020 AF488) or Alexa Fluor® 647 (sc-8020 AF647). Please contact Technical Service if you have further questions concerning this product.
