Cytokeratin 6 Antibody (B-7) is a mouse monoclonal IgG1 kappa light chain antibody that detects Cytokeratin 6 of human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). Anti-Cytokeratin 6 antibody (B-7) is available in non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Cytokeratin 6 belongs to the cytokeratin family, a diverse group of intermediate filament proteins essential for maintaining epithelial cell structural integrity. Cytokeratin proteins are expressed in pairs within keratinized and non-keratinized epithelial tissues, constituting up to 85% of mature keratinocytes in vertebrate epidermis. Cytokeratin 6 plays a vital role in cellular differentiation and tissue specialization, serving as a marker for tissue differentiation and characterization of malignant tumors. Cytokeratin 6 forms alpha-helical coiled-coil dimers that associate laterally to create 10 nm diameter filaments, providing mechanical resilience to epithelial tissues. Pro-inflammatory cytokines like interleukin-1 and tumor necrosis factor-alpha regulate Cytokeratin 6 expression, inducing transcription in epidermal keratinocytes through C/EBP β transcription factor. Multiple isoforms of Cytokeratin 6 (6A-6F) are encoded by highly homologous genes in humans, each showing distinct tissue expression patterns, with Cytokeratin 6A predominating in epithelial tissues. Human Cytokeratin 6A gene is located on chromosome 12q13, and mutations in this gene link to various inheritable hair and skin disorders, underlining Cytokeratin 6′s importance in dermatological health.
