Cytohesin-1/2 Antibody (D-11) is a mouse monoclonal IgG1 kappa light chain antibody that detects cytohesin-1/2 protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). Anti-cytohesin-1/2 antibody (D-11) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Cytohesin-1 and cytohesin-2, also known as ARNO (ARF nucleotide-binding-site opener), function as guanine nucleotide exchange factors (GEFs) for ARF (ADP Ribosylation factor) proteins, which are crucial for vesicle budding and trafficking within cells. The structure of cytohesin-1 and cytohesin-2 features an N-terminal coiled-coil domain, a Sec 7 domain that facilitates GDP/GTP exchange, and a C-terminal pleckstrin homology (PH) domain. The PH domain enhances the GEF activity of cytohesin-2 through interaction with phosphatidylinositol 4,5-bisphosphate, helping recruit cytohesin-2 to cellular membranes. This recruitment regulates ARF proteins, as cytohesin-1 catalyzes nucleotide exchange on ARF1 and ARF3, while cytohesin-2 stimulates nucleotide exchange on ARF1 and ARF6, thereby influencing various cellular processes such as membrane trafficking and cytoskeletal dynamics.
