SEE ALSO...

CYP3A4 Antibody (HL3) is a mouse monoclonal IgG1 kappa light chain antibody that detects Cytochrome P450 3A4 (CYP3A4) in human and rat samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and immunohistochemistry. Anti-CYP3A4 antibody (HL3) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. CYP3A4 plays a crucial role in drug metabolism, as CYP3A4 is responsible for the biotransformation of over 50% of clinically used medications, making CYP3A4 essential for pharmacokinetics and personalized medicine. Located predominantly in the endoplasmic reticulum of liver cells and upper intestinal enterocytes, CYP3A4′s expression can be induced by glucocorticoids and other pharmacological agents, which can significantly affect drug clearance and efficacy. The CYP3A gene cluster, located at the 7q22.1 locus, includes CYP3A4 along with CYP3A5, CYP3A7, and CYP3A43, as well as two pseudogenes, highlighting the complexity and importance of this enzyme family in drug metabolism and the synthesis of cholesterol, steroids, and other lipids. Understanding the function and regulation of CYP3A4 is vital for optimizing therapeutic strategies and minimizing adverse drug reactions.

Hello,I am working with HEPG2 cells, my bands are around 36 kDa. I think they are in the wrong place because there is no difference in different siRNA applications. I used odontoblast ( not expressed CYP3A4), but there is a band. Any recommendation?

Can you recommend a positive control to use with CYP3A4 Antibody (HL3): sc-53850 for Western blot?

Asked by: TinTin
We recommend human liver tissue extract (sc-363766) to use as positive control as we have Western blot data showing specific detection of CYP3A4 in this sample.

Rated 5 out of 5 by from specific signalI used this antibody detect CYP3A4 protein , it shows high specific signal was observed for CYP3A4 expressing in human liver tissue with no signal from negative control samples at dilution 1:600,the result is satisfied to our mind.
Date published: 2017-07-29
Rated 4 out of 5 by from western blotIn HepG2 cells, the signal is high and bands are clear, but the backgroung is also high at the dilution of 1:500, so we want to try again.
